2AB標(biāo)記的多糖標(biāo)準(zhǔn)品溶解方法:
我們2AB標(biāo)記的多糖是粉末狀態(tài),量比較少,差不多只有100pmol. 所以很多時(shí)候是看不出來的,這很正常。
CAB-XX-01 可以溶解到50ul 的水中,UPHLC分析上樣用5ul 即可。一次使用不完可以存放-20℃,再次復(fù)溶。

CN-XXX-10U標(biāo)準(zhǔn)品溶解于200uL水中,然后取 0.5uL用于MALDI分析。只需直接加水到小瓶中并漩渦10秒,確保水沿著管子內(nèi)壁流動(dòng),混合均勻。
For the CN-XXX-10U, I confirm it is dissolved in 200uL water and then use 0.5uL of it for MALDI. Just add water directly to the vial and vortex for 10 sec,
making sure that water is going around the inner wall of the tube.

The CLIBN-IGG consists of polyclonal IgG and thus has a high number of glycoforms and a high number of glycopeptides species.
This means that the signal is diluted by having this larger range of glycopeptide structures, each of which is present at a lower level than if the sample had been a single glycoform.
That is why they couldn’t see any signal from the CLIBN-IGG sample and they should get signal from the single glycoform standard.
Another reason is the CLIBN-IGG they bought might be the 5ug aliquot not 25ug (see CofA)
